چکیده

In this study, urease was isolated from Cicer arietinum and immobilized in calcium alginate beads. Various parameters, such as effect of thermal stability, temperature, optimum pH and pH stability, substrate concentration, reuse and storage stability were investigated and the results of the investigation were compared with the soluble enzyme. The activity yield of immobilization was calculated as 88.5 %. Optimum temperature and pH were found to be similar for both soluble and immobilized enzymes. It was observed that immobilization did not change pH and temperature prompt of the enzyme. pH and optimum temperature were found to be 7.0 and 50°C respectively. Improved thermal and pH stability of urease were achieved by immobilization. The Km value for immobilized urease was found to be higher than that of the free enzyme. Immobilization of beads at optimum conditions enabled up to 5 repeated use of enzyme and maintained 58% of their initial activity. It was found that storage stability of immobilized enzyme was better than that of the free enzyme. Immobilized urease enzyme was tested for the determination of urea amounts in various animal feeds.

Determination of Genetic diversity of cultivated chickpea (Cicer arietinum L.) using Medicago truncatula EST-SSRs

Expressed sequence tags simple sequence repeats (EST-SSRs) are important sources for investigation of genetic diversity and molecular marker development. Similar to genomic SSRs, the EST-SSRs are useful markers for many applications in genetics and plant breeding such as genetic diversity analysis, molecular mapping and cross-transferability across related species and genera. In spite of low po...

A Conductometric Urea Biosensor by Direct Immobilization of Urease on Pt Electrode

Urease was immobilized on platinum electrode both by chemical binding and electropolymerization. The conductometric urea biosensor thus prepared showed a detection limit of 4.9×10-5 M and linear dynamic range from 4.9×10-5 to 5.8×10-3 M for urea concentration when the enzyme is covalently immobilized on Pt electrodes. Conductometric transducers respo...

Utilization of Urea and Urease Treated Rice Straw by Indigenous Growing Cattle

The experiment was conducted to study the effect of treatment of rice straw with urea and a urease containing midden soil on the chemical composition of treated rice straw, feed intake of the animals, nutrients digestibility, body weight gain, feed conversion efficiency and overall economy of feeding for a period of 105 days. Twelve indigenous growing cattle (live weight 130.00±1.67 kg) were se...

Flow Injection System with Potentiometric Detection for the Determination of Urea Content in Milks

Two variant manifolds of a flow injection analysis (FIA) system are described for the determination of urea content in milks. This determination consists of the enzymatic reaction of urea with urease in which ammonium ion is formed. Ammonium is converted to ammonia by adding a NaOH solution and then led to a gas diffusion unit in which it diffuses to an acceptor channel (Tris/HCl, pH 7.5). Here...

Anticonvulsant Effect of Cicer arietinum Seed in Animal Models of Epilepsy: Introduction of an active Molecule with Novel Chemical Structure

Background: Cicer arietinum (Chickpea) is one of the most important harvests in the world with high nutritional value. Lack of essential oils in the seeds of Chickpea is an advantage in search for drug-like molecules with less toxicity. We evaluated anticonvulsant effect of C. arietinum in common animal models of epilepsy. Methods: Dichloromethane extract was obtained from C. arietinum seeds by...
